النَّهَارِ — of the day. This is the owner in the 'middle of ___' pairing, so it sits in the genitive. The word picks out the daylight span specifically, as opposed to the night; Arabic keeps separate words for the two halves.

And seeking help from the Almighty God during the night and the day,
وَالنَّهَارِ — and the day. This is the bound wa- 'and' plus al- 'the' plus a noun, 'and the day', coordinated onto 'the night'. The wa- joins it on equal footing, and because it parallels 'the night' it shares the same possessed ending, rounding out 'night and day'.
