وَجَعَلَ سُبْحَانَهُ الإِمَامَةَ فِي الدِّينِ مَنُوطَةً بِالصَّبْرِ وَالْيَقِينِ
And He, exalted be He, made leadership in religion dependent on patience and certainty.
الدِّينِ — the religion. Made definite by al- and pulled into the genitive by the preposition before it. The case ending is purely the preposition's doing; the al- marks 'the religion' as a known, specific sphere.

So he made all of this part of the religion.
الدِّينِ — the religion. A definite noun 'the religion' governed by 'of' into the genitive, the category the matter is made part of. The preposition forces the genitive ending, completing 'part of the religion'.
