وإذا كانت جميع الحسنات لا بد فيها من شيئين
And if all good deeds must include two things:
الحَسَنَاتِ — good deeds. The owner completing 'all of the good deeds', in the genitive. On this sound feminine plural the genitive and accusative share one ending, so context, not the vowel alone, fixes its role.

It requires good understanding of interpretation and an abundance of stories about the poor and ascetics.
حَسَنَةً — good. This adjective follows its noun and matches it — feminine, indefinite, accusative — telling you it belongs to 'understanding', not to anything else in the sentence. Arabic relies on this echoing of gender and case to bind an adjective to the right noun.
