Quraysh realized that his companions had increased in number.
قُرَيْشٌ — Quraysh. The subject of 'knew', nominative (the tanwin), after its verb — the Meccan tribe. A tribal name treated grammatically as feminine.

So he would spend the morning in Mecca with the Quraysh, sitting together in small gatherings.
قُرَيْشٍ — the Quraysh. A tribe name. As a proper name it is treated as already definite, so it takes no 'the'; standing after 'with', it fills the slot of the people he keeps company with and sits in the 'of' ending the preceding word demands.
