فَتَجِدُ أَحَدُهُمْ يَسْأَلُ عَنْ الْأَمْرِ الصَّغِيرِ وَالْكَبِيرِ،
So you find one of them asking about small and large matters.
الْأَمْرِ — the matter. Carries al- ('the'), making it specific, and is the object of the 'about' preposition before it, so it takes the genitive ending. It names the matter asked about, which the following adjectives will then stretch from small to large. The definiteness treats it as the topic in view.

And Allah has commanded in His Book obedience to Him, obedience to His Messenger, and obedience to those in authority among the believers.
الْأَمْرِ — the authority. The 'the' makes this noun definite, and it closes the 'of' pairing as the owner: 'possessors of the command'. Standing second in that pairing it takes the genitive ending; together the two words name the people in charge.
