قَلْبٍ — heart. The owner in 'work of ___', in the genitive but left indefinite, so the pairing means heart-work in general rather than 'the heart' specifically. An 'of' pairing stays indefinite when its final owner is indefinite.

قُلُوبِكُمْ — your hearts. This is 'hearts' with 'your (plural)' attached, in the preposition's object form. The plural noun and the plural-you possessor are bundled into one word; this is the inner thing God is said to regard.
